Estuarine molecular bycatch as a landscape-wide biomonitoring tool

Environmental DNA analysis is rapidly transforming biodiversity monitoring and bolstering conservation applications worldwide. This approach has been assisted by the development of metabarcoding PCR primers that are suited for detection a wide range taxa. However, little effort gone into exploring value non-target sequences generated in every survey, but subsequently discarded. Here we demonstrate fish-targeted markers widely employed aquatic biomonitoring can also detect birds mammals present surrounding habitats. We showcase this feature three temperate estuaries over multiple seasons, where dozens bird mammal species offer valuable insights spatial temporal faunal variation. Our results indicate existing metabarcode sequence data sets suitable mining exploration ‘molecular by-catch’, future eDNA-based surveys be designed to accommodate enhanced property applicable tool. • used an eDNA survey UK coastal fishes specifically search non-fish taxa; Nearly 18% reads mapped 50 mammals; analysed spatially across estuaries, found patterns relevance; These taxa detected, here termed “molecular bycatch”, represent important source information.
